Many large studies have implemented wrist or thigh accelerometry to capture physical activity, but the accuracy of these measurements to infer activity energy expenditure (AEE) and consequently total energy expenditure (TEE) has not been demonstrated. The purpose of this study was to assess the validity of acceleration intensity at wrist and thigh sites as estimates of AEE and TEE under free-living conditions using a gold-standard criterion. Measurements for 193 UK adults (105 men, 88 women, aged 40–66 years, BMI 20.4–36.6 kg m−2) were collected with triaxial accelerometers worn on the dominant wrist, non-dominant wrist and thigh in free-living conditions for 9–14 days. In a subsample (50 men, 50 women) TEE was simultaneously assessed with doubly labelled water (DLW). AEE was estimated from non-dominant wrist using an established estimation model, and novel models were derived for dominant wrist and thigh in the non-DLW subsample. Agreement with both AEE and TEE from DLW was evaluated by mean bias, root mean squared error (RMSE), and Pearson correlation. Mean TEE and AEE derived from DLW were 11.6 (2.3) MJ day−1 and 49.8 (16.3) kJ day−1 kg−1. Dominant and non-dominant wrist acceleration were highly correlated in free-living (r = 0.93), but less so with thigh (r = 0.73 and 0.66, respectively). The biochemical and organoleptic characteristics of the longissimus thoracis muscle from suckler bulls (n = 56) finished on a concentrate-based system (C) or raised in a pasture-based system (P) incorporating 99 (P99), 162 (P162) or 231 days (P231) of grazing prior to indoor finishing on the concentrate-based diet were investigated. Age at slaughter increased with increasing period at pasture. Intramuscular fat concentration was lower (p < 0.001) for P99 than for C, P162 and P231 bulls, which did not differ. Soluble collagen proportion was lower (p < 0.01) for P162 and P231 than for P99 and C bulls. Collagen cross-link content was higher (p < 0.05) for P231 than for P99 and C bulls and for P162 than for C bulls. The proportion of type I muscle fibres was higher (p < 0.01) for P231 and P162 than for P99 and C bulls. Sensory tenderness was higher (p < 0.001) for C and P162 than for P99 and P231 bulls and overall liking was higher (p < 0.01) for C than for P99 and P231 bulls but similar to P162 bulls. Recent technological advances have transformed the research on physical activity initially based on questionnaire data to the most recent objective data from accelerometers. The shift to availability of raw accelerations has increased measurement accuracy, transparency, and the potential for data harmonization. However, it has also shifted the need for considerable processing expertise to the researcher. Many users do not have this expertise. The R package GGIR has been made available to all as a tool to convermulti-day high resolution raw accelerometer data from wearable movement sensors into meaningful evidence-based outcomes and insightful reports for the study of human daily physical activity and sleep. This paper aims to provide a one-stop overview of GGIR package, the papers underpinning the theory of GGIR, and how research contributes to the continued growth of the GGIR package. The package includes a range of literature-supported methods to clean the data and provide day-by-day, as well as full recording, weekly, weekend, and weekday estimates of physical activity and sleep parameters. In addition, the package also comes with a shell function that enables the user to process a set of input files and produce csv summary reports with a single function call, ideal for users less proficient in R.
